Abstract

[Objective] To establish a two-dimensional polyacrylamide gel electrophoresis( 2-D PAGE) system for suspension cell proteomic analysis of Betula platyphylla. [Method]the improved acetic acid( TCA)-acetone precipitation method was applied for suspension cell protein isolation and purification,and the key technologies of electrophoresis including the sample loading quantity and types of IPG strip were optimized. [Reault]The results indicated that the reproducible 2-D gel with more spots and high resolution electrophoresis diagram could be obtained by using the improved TCA-acetone precipitation method,with 9. 0 μg / μl of loading protein sample on 17 cm IPG strip with p H 4-7. About 1 000 protein spots were obtained by PDQuest software analysis. 3 randomly selected protein spots were used to identify by matrix assisted laser desorption ionization time of flight mass spectrometry( MALDI TOF MS). The identified 3 proteins were S-S-adenosylmethionine synthetase,transposase for IS1330 and transcriptional regulatory proteins of PadR family,respectively. [Conclusion] The study laid the foundation for using proteomics analysis inducer to improve secondary metabolites accumulation mechanism.
