Objective To investigate the influence of memantine hydrochloride on the cognitive function and inflammatory factors expression of patients with Parkinson's disease with dementia(PDD),and explored its related mechanism.Methods A total of 72 cases with senile PDD which were treated in authors' hospital was divided into the experimental group and the control group with a completely randomized method.The experimental group was given memantine hydrochloride treatment,the control group was given odd donepezil hydrochloride treatment.Before treatment,4 weeks,8weeks,16 weeks after treatment,the patients' interleukin 1β(IL-1β),IL-2,IL-6,tumor necrosis factorα(TNF-α),interferonγ(IFN-γ)inflammatory cytokines were detected.The inflammatory cytokines in 36 healthy people were tested.Montreal cognitive assessment(MoCA)was used to assess cognitive function.The occurrence of adverse reactions were observed.Results There was no significant difference in MoCA scores of two groups patients before treatment(P0.05).After treatment MoCA scores were significantly higher in two groups(P0.05),while the difference between the two groups were statistically significant(P0.05)and MoCA score was higher in the experimental group.The inflammatory factors were significantly higher in the experimental group and the control group than healthy people(P0.05).After treatment the inflammatory cytokines in the control group was no significantly difference(P0.05),while IL-1β,TNF-αin the ex-perimental group was significantly lower(P0.05),IL-2,IL-6,IFN-γ did not change significantly difference(P0.05).Multiple linear regression analysis showed that IL-1β,TNF-α,age,medication had linear relationship with MoCA scores(P0.05).Conclusion The inflammatoryresponse plays an important role in the pathogenesis of Parkinson's disease,and is closely related with the cognitive function of patients,memantine hydrochloride improve cognitive function by inhibiting local and systemic inflammatory response.
