Objective:To obtain optimal conditions of lettuce transgenetic system,which could provide the condition for the further transformation study of transgenic plant expression vector against dental caries.Method:By using the six different varieties lettuce as explants,the technology of plant regeneration was studied.Several factors such as bud ratio,hormone concentration and the antibiotic selection pressure and so on,were investigated to optimize the regeneration system in vitro.Result:The experimental results showed that compared with six different varieties of lettuce,The seed of Yi Dali lettuce sprout fastest and its bud ratio is the highest.It is shown that the best optimal conditions is that the 3-4 day old lettuce plant cotyledon grew on the 1/2 MS+0.05 mg/L NAA+0.1 mg/L 6-BA medium;It was optional to choice 50 mg/L paromomycin as the antibiotic selection pressure.Conclusion:The conditions of lettuce transgenetic system,which should provide to transformation of transgenic plant expression vector against dental caries,were successfully constructed.
