Abstract
Objective:To investigate the traffic of HTLV-1acrossing the bl oo d-brain barrier and viral entring into the CNS,in order to indicate the mechani sm of HAM/TSP.Methods:CSF mononuclear cells and peripheral blood mononuclear cel ls(PBMC)were collected from5patients with HAM/TSP.The amplification of HTLV-1i ntegration sites between CSF and PBMC was compared by Linker-mediated polymerase chain reaction(LMPCR).Results:Such oligoclonal distribution of HTLV-1-infecte d cells was evidenced in the cerebrospinal fluid(CSF)derived from5patients wit h HAM/TSP.Furthermore,clonal populations of HTLV-1-infected lymphocytes sharing the same HTLV-1proviral flanking sequences(i.e.,integration sites in the cell ular DNA)were found in both the CSF and the peripheral blood.Conclusion:HTLV-1c rosses the blood-brain barrier by migration of HTLV-1-infected lymphocytes in vi vo.The proliferation of infected cells will lead to pathogenic mecha-nisms of H AM/TSP.
