Influence of tripterygium po1yglycoside on free radicals and inflammatory factors in rats with adjuvant arthritis

Abstract
Objective To investigate the protective effect and mechanism of tripterygium po1yglycoside (TII) on rats with adjuvant arthritis (AA). Methods The rat AA model was established and 30 SD male rats were divided randomly into the normal group, model group and TII group. The swollen of rat claw were measured, the histological changes were observed by the light microscope, the activity of superoxide dismulase (SOD), MDA level and T-AOC were detected by the biochemical method, the contents of TNF-α and PGE2 in claw were detected by the radioimmunoassay, and the expressions of COX-2 and NF-κB in cartilage of knee joint were detected by immunohistochemistry. [WTHZ]Results Compared with the normal group in the model group the activities of SOD and T-AOC were decreased (P0.01), MDA level was increased, the contents of TNF-α and PGE2 were increased, and the expressions of COX-2 and NF-κB were increased. In the TII group the activities of SOD and T-AOC were increased, the MDA level, the contents of TNF-α and PGE2, and the expressions of COX-2 and NF-κB were decreased. The result of the light microscope showed that in the TII group the hyperplasia of synovial membrane and inflammatory infiltration was relieved. Conclusion Tripterygium po1yglycoside has a protective effect on rat AA model. One of its mechanisms may be related to its antioxidation and inhibitory effects on inflammatory infiltration.
