The ultrastructure of polysaccharide isolated from the root of Paeonia suffruticosa Andr named as PSM2b-A was examined by transmission electron microscopy.PSM2b-A samples in aqueous solution were heated at 45℃ for 20 minutes before dipping to the copper grids following negative staining by uranyl acetate after air-dried.Coiled balls or circles,extended chains,network-like structures have been observed.It is presumed that polysaccharide molecules of PSM2bA in aqueous solution are likely to coil into floppy or compact globular structures which may assemble into aggregates by means of hydrogen bonds,leading to a variety of morphology.The TEM images provide useful insight into the conformations and interactions of polysaccharide molecules.
