Abstract
Germination of Sophora's seeds was limited by the thick skin.For improving germinant rate,the seeds were exposed to concentrated sulfuri aid for 30,60,90 and 120 minutes or hot water for 90,120 and 50 minutes or seed skin was removed.Results showed that germination rate increased at first,and then decreased.The optimal treatment was exposed to concentrated sulfuri aid for 90 minutes and germination rate was higher(83%) than that of others treatment.When the germinated seeds were treated with 0.01%,0.1% or 0.2% colhiines for 48,72 or 108 h,respetively.Exposure to 0.2 % colchicines for 108 h produced polyploid-induced percentages(68%).Exposing to 0.2 % colchiines for 48 h led to the lowest rate(8%).Further,the most efficient treatment to the root tip cells was with 0.2% colchicines for 72 h,after being treated 48 h in 0.1 % colchicines occured the exact opposite phenomenon.So far,the chromosome-doubled rates of Tetraploids was much higher than Octploid,meanwhile,the mixoploidic was observed during this period.
