Bioassay of Hyphantria cunea Nuclear Polyhedrosis Virus Isolated from Qingdao

Abstract
The bioactivity of Hyphantria cunea nuclear polyhedrosis virus(HcNPV) isolated from Qingdao was tested on fourth,fifth and sixth instar larvae of Hyphantria cunea.The bioassays showed the median lethal concentration(LC50) for fourth,fifth and sixth instar larvae were 1.99×105,1.69×106 and 8.04×106 occlusion bodies(OBs)/ml,respectively.When larvae were fed on diet with virus concentration of 1.48×107 OBs/ml,the median lethal time(LT50) values for fourth,fifth and sixth instar larvae were 6.14,8.63 and 10.31d,respectively.When larvae were fed on diet with virus concentration of 1.48×108 OBs/ml,the LT50 values for fourth,fifth and sixth instar larvae were 5.30,5.92 and 7.12d,respectively.The LT50 values for 1.48×106,1.48×107 and 1.48×108 OBs/ml were 7.65,6.14 and 5.30d,respectively,in fourth instar assay.The lethal time increased with increased larval instars,whereas the relationship between the LT50 and the fed-virus concentration was reversed.The bioactivity determination offered potential of this indigenous HcNPV in management of Hyphantria cunea.
