A Prelimary Study on the Transformation of Glycyrrhiza uralensis Genome DNA in Hetian Alfafa Implanted by Argon Ion

Abstract
The total DNA of Glycyrrhiza uralensis were randomly implanted into Medicago sativa(Hetian alfalfa) by argon ion,and induced obvious biological effects on the normal growing and cultivating of the alfalfa.The seed-setting rate of T 1 generation transgene alfalfa was only 11.66%,and normal seeds was 64.29%.The qualitative test of the natural products was carried out to the nine individual plant's roots and stems of T 1 generation transgene alfalfa with well developed seeds.The results of natural products detection showed that some saponins were contained in roots of the two individual plants.X-ray diffraction was used to scan medicinal transgenic alfalfa and the diffract gram map were obtained.The polysaccharide of control group and T 1 generation were extracted from roots and stems and determined.The results showed that after searching peaks-processing,the relative error was 0.67,the difference of crystal spacing was greater than the allowed error 0.02,which reflected that the micro-structure or component of the sample changed.The contents of polysaccharide in the transgenic alfalfa's stems and roots were 0.890%,4.815% respectively,compared with that of the common alfalfa's 1.064%,4.882%.These results reflected that the content of polysaccharide in transgene alfalfa decreased which implanted Glycyrrhiza uralensis total DNA.
