Abstract
Surface sterilization is the key step in isolation of endophytes from plants. We explored a new suface sterilization method to isolate endophytic fungi from Taxus chinensis var. mairei with 75% ethanol and 2% sodium hypochlorite( Na Cl O)instead of mercuric chloride. After pretreated by immersing in 75% ethanol for 3 min,the yew leaf,stem and bark were all immersed in 2% sodium hypochlorite solution for 2 min,and then immersed in 75% ethanol for 3 min,5 min and 7 min,respectively. The method has a good surface sterilization effect.
