Abstract
We tested for the proper concentrations of template,primer,dNTP and enzyme by an orthogonal design experiment.The result showed that the difference between genome did not obviously affect the efficiency of asparagus bean SSR amplification.The further analysis revealed that only the primer concentration was the key factor for the efficiency of SSR amplification.Taking together cost of SSR analysis and the results of the following experiments for primer concentration,we determined the optimal concentrations for asparagus bean SSR amplification.Namely,the concentrations of primer,template,dNTP and Taq polymerase were 1.6 μmol/L,1 ng/L,100 μM/L and 0.5 U,respectively.The SSR primer concentration were discussed in the paper.This study set up a foundation for the genetic analysis of asparagus bean with SSR technique.
