To observe the effect of vasoactive intestinal peptide (VIP) on intestinal lymphocytes homing at gut-associated lymphoid tissues (GALT) in rats with multiple organs dysfunction syndrome (MODS),the superior mesenteric artery was occluded for 45 minutes and reperfused for 6 hours to make the model of MODS. VIP was infused into rats from femoral vein and from abdomen. Intestinal lymph was collected to measure the population of lymphocytes per hour and the percentage of T and B cells. In addition,the intestinal lymphocytes from intestinal lymph were labeled with 51 Cr and then infused into femoral vein of rats with MODS. 51 Cr-intestinal lymphocytes in intestinal mucosa and Peyer's patches were counted with γ counter. The level of endotoxin and TNF-α in intestinal lymph and D-(-)-lactate acid,ALT,cretinine,PaO 2 in plasma was determined,and the histology of essential organs was compared between MODS and MODS treated with VIP. The results showed that VIP increased the population of intestinal lymphocytes [(0.42±0.180×10 7/h vs (0.28±0.15)×10 7/h, P 0.05] trafficked from intestinal mucosa into blood circulation in rats with MODS. The percentage of 51 Cr-intestine lymphocytes in Peyer's patches (2.14%±1.49%) and intestine mucosa (1.58%±0.42%) of rats with MODS treated with VIP was remarkably decreased in compared with those of rat with MODS (5.04%±1.23%, P 0.01,3.23%±1.69%, P 0.05). The endotoxin or TNF-α level of intestine lymph and D-(-)-lactate acid,ALT and cretinine in plasma of rats with MODS was greatly decreased after in fusion of VIP PaO2 was increased to 4 9%,and the severe inflammation in the essential organs was relieved It condudes that the decrement of endotoxin or TNF αand improvement of the function of each organ might be related with decline of intestinal lymphocytes homing to GALT caused by VIP that relieved the severe inflammation in intestinal mucosa
