Electrophysiological Effects of Zhigancao Decoction on Guinea Pigs with Ischemia-hypoxia-induced Arrhythmia

Abstract
Objective To explore the electric activities of left ventricular outflow tract induced by hypoxia and ischemia, and to investigate the antiarrhythmic effect of Zhigancao Decoction. Methods Conventional intracellular microelectrode technique was used to record the electrophysiological parameters of spontaneous slow action potential duration (APD), 50 % of APD (APD50) and 90 % of APD (APD50), automatic depolarization velocity (VDD) and rate of potential frequency (RPF) in guinea pigs left ventricular outflow tract pacemaker cells of normal control group, ischemia- hypoxia group and ischemia- hypoxia adding Zhigancao Decotion group. Results Compared with normal controls, action potentials of APD50, APD90 and APD of ventricular outflow tract in ischemia- hypoxia group were significantly shortened (P 0.05), VDD and RPF significantly slowered (P 0.01), and there existed arrhythmia .The addition of Zhigancao Decotion can significantly prolong APD50, APD90 and APD, accelerate VDD and RPF (P 0.05). Twenty minutes after reperfusion, spontaneous discharge frequency recovered to the normal control rhythm. Conclusion The result indicates that the automaticity of left ventricular outflow tract can be influenced by hypoxia and ischemia, and Zhigancao Decoction can counteract arrhythmia induced by ischemia and hypoxia, suggesting that the therapeutic mechanism of Zhigancao Decoction for the treatment of arrhythmia induced by ischemia and hypoxia is related to the regulation of the slow response of left ventricular outflow tract abnormalities.
