Abstract
Callus induction regeneration system of wheat,genetic transformation of wheat using and agrobacterium-mediated approach were applied in the research to study the genetic transformation conditions of wheat mediated by agrobacterium tumefaciens.The factors affecting the transformation,such as pre-cultured,co-cultivation,the selective pressure of antibiotic,Cef on the regeneration,agrobacterium concentration and infection periodwere examined.The optimized procedure was as following:after pre-cultured for 1 to 2 days,the callus induction were dipped into an a agrobacterium which concentration was OD600 0.6 for 15min,then co-cultivation for 3 days,the explants were transferred to selective media with 100mg/L Km and 500mg/L Cef.These conditions were benefited to improving the transformation efficiency.PCR analysis showed that objective gene were transferred to wheat genome.
