Research of the expressing of iNOS and HSP70 genes induced by IFNγ in isolated swine heart

Abstract
Objective To research the effects of interferon γ(IFNγ) on the two genes′expressions of iNOS and heat shock proteins 70 (HSP70),and on the long term protection of the preserved swine heart induced by late precondition. Methods Thirty swine were preconditioned by subcutaneous injection respectively and divided randomly into three groups : with normal saline, with IFNγ , and with IFNγ + aminoguanidin. After the basic measurements of left ventricular function, LDH and CK MB leakage, the hearts were isolated and kept in 4℃ cold crystalloid liquid. After 4 hours reservation, the isolated hearts were reperfused with warm blood ,and rebeated automatically or by electric shock. The samples of myocardial tissue were taken into liguid nitrogon at 60 min for measuring the expression of the two genes by RT PCR and Wstern blot. Results Compared to β actin, the HSP70 expression by RT PCR and Western Blot was evidently up regulated in two experience groups, the iNOS expression was evidently up regulated in IFNγ group, but not in IFNγ+ aminoguanidin group. Conclusion IFNγ could increase markedly the expression of iNOS and HSP70i genes, with positive influence on isolated swine heart.
