Extracting DNA from young leaves of sweet potato,and the purity,concentration of the DNA were tested.The results showed that the DNA could be well amplified,which was fully able to meet the demand of further experiments.At the same time,impacting factors of sweet potato ISSR-PCR reaction system were optimized,and established the best suitable reaction system for sweet potato.That was in 20 μl reaction system,DNA template with 10 times of dilution concentration,0.30 mmol/L dNTP,1.00 μmol/L primerand 1.00 U Taq enzyme and 0.5% isopropanol.
