For further improving the efficiency of anther callus induction,twelve F1 hybrids of winter wheat from Huang-Huai wheat areas were used for anther culture.The effects of different basal media,culture methods,anther density and the number of co-culture ovaries were examined on wheat anther callus induction and differentiation.The results showed that:1.The CHB medium had the best effect on anther callus induction,the C17 medium took the second place,while the N6 medium as well as the W14 medium had poor effects,and the different genotypes had various response at the different media,but most of the genotypes on the CHB and C17 medium had a better response.2.Low concentration of 2,4-D(0.5 mg/L) was favourable to forming embryoid and direct regeneration plantlet on the induction medium compared with the higher 2,4-D concentration(2.0 mg/L).3.When anther cultured in liquid medium,the callus induction rate was the highest,but the differentiation of the callus was inhibited,the callus induction rate was the lowest in solid medium and double layer medium,while the induction and regeneration of callus was better by semisolid medium culture which was a better culture method for wheat anther culture.4.Within experimental anther density,our results indicated that the inclusion of ovaries in the induction medium significantly increased the induction efficiency of anther calli in two winter wheat genotypes tested.When the anther inoculation density was low(2-2.4 anthers per mL),about 20 ovaries per culture bottle could obviously promoted inductive effect of calli,while the anther inoculation density was middle or high(4-4.8 or 6-7.2 anthers per mL),more than 10 or 10 ovaries had a better induction effect.
