Genotype is the main limiting factor to improve the Agroacterium-mediated wheat transformation efficiency.Twelve Chuannong wheat cultivars were evaluated for the susceptibility to Agrobacterium,by use of strain AGL 1 carrying PCAMBIA-1303.The results from GUS gene transient expression assay showed that transformation rate was quite different among these genotypes.Using immature embryo as explant,the transformation rate of Chuannong 21 and Chuannong 22 were much higher than others,up to 90% and 85% respectively.And the transformation efficiency of callus generally higher than that of immature,among which transformation rate of Chuannong 22 and Chuannong 25 both reached 100%.So,these cultivars could be applied for useful gene transformation.
