Abstract
Objective:To explore the effect of the Rou-Bin manipulation to the apoptosis and ultrastructure of cartilage cell with rabbit's knee joints,and the mechanism of Rou-Bin manipulation to the knee joint cartilage tissue of the rabbits.Methods:50 rabbits were randomly divided into manipulation group,injection group,model group,dummy-model group and normal group,10 for each group.ligaturing and cutting off the inferior-gluteal vein,femoral vein and great saphenous vein on the right side of the rabbit for the first 3 groups,shows the same veins without cutting off for dummy-model group,no treatment for the normal group.1 week after modeled,17 times of Rou-Bin manipulation on each rabbit in manipulation group,the rabbit injection with sodium hyaluronate 5 times in inject group,no special treatment for other rabbits.The 8th weekend after modeled,to take all the rabbit right knee medial tibial plateau cartilage tissue processing,detected chondrocyte apoptosis rate by TUNEL,to observe the change of the ultrastructure of cartilage cell by TEM,statistics processing.Results:Cartilage degeneration clearly for the model group,chondrocyte apoptosis rate is 17.25±3.24%,hint by TEM:many apoptosis chondrocyte;Cartilage degeneration clearly for dummy-model group,chondrocyte apoptosis rate is 16.63±2.13%;Cartilage degeneration lightly for the manipulation group and inject group,chondrocyte apoptosis rate is 8.40±2.84% for manipulation group,hint by TEM:good structure with the cell;chondrocyte apoptosis rate is 9.50±3.25% for inject group,hint by TEM:good structure with the cell;no cartilage degeneration for the normal group,chondrocyte apoptosis rate is 3.88±1.23%,hint by TEM:the structure with the cell nearly normal.To compare with model group,all proportions of the chondrocyte apoptosis rate for the manipulation group,inject group and normal group are 0.01,statistically significant difference.Conclusion:The Rou-Bin manipulation can help the apoptosis of cartilage cell with rabbit's knee joints lower,to keep the structure of cartilage well.
