Synthesis of Metallothionein in Rat Aortic Vascular Smooth Muscle Induced by Low Power Laser

Abstract
Objective To study the effect of low power laser on the synthesis of metallothionein in the cultured rat thoracic aorta for further study of the mechanism of low power laser's cell protection function. Methods Rat thoracic aorta was cultured in Krebs-Henseleit solution containing 10% FBS and was irradiated with low power red laser (632 nm) at doses of 3, 6 and 12 J/cm 2, respectively. Then the mRNA level of metallothionein in the thoracic aorta was studied by reverse-transcription competitive PCR. And the secretion of metallothionein in the thoracic aorta was measured with 109 Cd and bovine hemoglobin. Results The secretion of metallothionein was increased by 142.8%(P0.05), 170.8%(P0.05)and 142.1%(P0.05) in thoracic aorta irradiated with 3, 6 and 12 J/cm 2, respectively. And the metallothionein 1 mRNA levels were increased by 175%(P0.05), 418.6%(P0.05) and 668.2%(P0.01)compared with control at doses of 3, 6 and 12 J/cm 2, respectively, while metallothionein 2 mRNA levels were increased by 163.9(P0.05), 615%(P0.01) and 1 087%(P0.01), respectively. Conclusions Low power laser can increase metallothionein both in gene transcription and protein level.
