Abstract
Objective:To develop differentiation and plant regeneration system from in vitro leaf tissues of Chirita medica D.Fang ex W.T.Wang,an endangered medicinal species.Methods:The leaf of Chirita medica as explants were cultured on MS supplemented with different kinds and concentrations of hormone to select optimized different phase.Results:The optimum medium for adventitious buds induction and seedling regeneration was MS+1.0 mg/L 6-BA+0.2 mg/L NAA(pH 6.0) and the induce rate of bud regeneration was 100%.The efficient medium for high regeneration frequency was MS+0.5 mg/L 6-BA+0.2 mg/L NAA(pH 6.0) and the proliferation coefficient was 12.6/30 d.The optimal rooting medium was 1/2 MS+0.2 mg/L NAA(pH 6.0) and the rooting rate was 100% and the rate of transplanting survival was 95.0%.Conclusions:This system could be used to develop more seedlings in short time,and to provide guidance for conserving and sustainably developing the resource.
