Abstract
In this paper,the main indices of ISSR were studied taking the genomic DNA extracted from Bromus ciliatus L.as template.The optimum reaction system and reaction process of ISSR were found.The results showed that the reaction system was 20 μl.It included 2.0 mmol/L Mg2+,2.0 U Tap enzyme,0.3 mmol/L dNTP,0.5 μmol/L primer,30 ng DNA template.The reaction program was devised for 5 minutes at 94 ℃ in the first circle,45 seconds at 94 ℃ for 45 cycles,54 ℃ for 60 seconds,72 ℃ for 90 seconds,and 72 ℃ for 7 minutes in the final cycle.
