Abstract

The thymosin gene (thy) was introduced into lettuce by co-culturing excised cotyledon explants withAgrobacterium tumefaciens. The best transformation efficiency was obtained when explants were co-cultured on theselective medium MS1-1(MS+0.1 mg L-16-BA+0.05 mgL-1NAA+Cb500 mgL -1) containing 75 mgL-1 kanamycin. Roots wereinduced on MS1-2 (1/2MS+Kan50 mgL-1+Cb100 mgL-1). Integration of thymosin gene into the genome of lettuce plants wasconfirmed by PCR and Southern blot analysis. Expression of thymosin gene in transcriptional level was proved by RT-PCR.
