Abstract
Epoxy-modified acrylic have an epoxy group,which can react with an amino,a nucleophilic group on the protein,under mild conditions to achieve its immobilized.This paper studies the optimization of modified ionic exchanging resin on immobilization of laccase and enzymatic characterization.The results show that the immobilized enzyme activity is best under such conditions:the ammonium sulfate concentration 1.5mol·L-1,the amount of enzyme 0.2U·g-1,pH 6.5,temperature 35℃,fixed 3hours.At this time,vitality specific activity and activity recovery are 0.026 4U·g-1and 3.67%.The optimal conditions of immobilized enzyme catalyzed chemical reaction are as follows:temperature 45 ℃,pH 4.5,Km 1.233×10-3 mmol·L-1.The immobilized enzyme activity was inhibited with iron and calcium ions between the 4×10-3 to 4×10-4 moL·L-1,is activated between the 4×10-5 to 4×10-6 mol·L-1.
