Abstract
Separation of high-quality protoplasts is the foundation of study on carrying out cell functions and bi-ofilm characteristics.And it is an important condition for obtaining hih-yield and quality protoplasts to establish appropriate combination of concentration and digestion time and enzyme solution osmotic pressure.In this experiment,the impact of protoplast isolation of different enzyme concentration,the solution osmotic pressure and digestion time was studied by horseradish leaves as material.The results showed that:at 28.5℃ condition,when mixed enzyme solution composed of 1% cellulase+0.1% pectinase+0.9mol/L mannitol digested 50min at pH5.8,a higher protoplast yield and higher protoplast viability was received.
