Abstract
The phenol-sulfuric acid method were optimized with the factors affecting the determination of polysaccharide content in pumpkin seeds,including determination wavelength,temperatures, developing time,sulfuric acid amounts,and phenol amounts.Precision,accuracy and stability of this method were also discussed.Results showed that under the conditions of detection wavelength 486nm,phenol content 1.0mL,water-bath temperature 30℃ and developing time 30min,the accuracy and precision were good,with the average recovery 100.75%and the RSD was 0.83%.This method is simple and accurate.It will lay a foundation for further activity and function study on polysaccharides in pumpkin seeds.
