Abstract
The present paper describes an effective approach for the isolation of hypericin from the commercial dried extract of St.John's wort(Hypericum perforatum L.),which initially prepared by solid-phase extraction on polyamide and further separated and purificated with one-step process by high-speed counter-current chromatography(HSCCC) by using n-hexane-ethyl acetate-methanol-water(3∶5∶3∶5,v/v/v/v) as the two-phase solvent system.The experiment yielded 13.6mg of hypericin from 300mg of crude extract in one-step separation with the purity of 95% as determined by high-performance liquid chromatography(HPLC).The structures of the isolated compound was identified by 1H nuclearmagnetic resonance(NMR) and 13C NMR.
