The COPI vesicle associated protein Arf1 GTPase‐activation protein 1 (ARFGAP1) is required for lipid droplet biogenesis

Abstract
Excess free fatty acids are stored in liver as triglycerides in organelles called lipid droplets (LDs). They are not inert storage containers, but have proteins directed to their surface. This study identified ARFGAP1 on LDs derived from liver and cultured cells. ARFGAP1 regulates the dynamic cycling of ARF1 on Golgi membranes affecting COPI vesicle formation, SNARE priming and lipid metabolism. ARFGAP2 and 3 are also candidates for regulating ARF1 function on the Golgi membrane but have not been studied within the context of COPI function until recently. Previous studies on LDs showed ARFGAP1 responds to changes in diacylglycerol, implicating it in triglyceride synthesis the main component of LD. We show a requirement for ARFGAP1 in LD biogenesis using a combination of proteomics, siRNA, immunolocalisation techniques and GFP fusion proteins. We will discuss the possible machinery in which ARFGAP1 is involved in regards to LD production and/or fusion.
