Several routes for the manufacture of novel bacterial topo isomerase inhibitors (1, 2) [1] at Actelion Pharmaceuticals Ltd are presented (Fig. Whereas Route 1, comprising more than 30 chemical steps, was developed to rapidly manufacture the Ac tive Pharmaceutical Ingredients (API) for first clinical studies, Route 2 was designed and developed to cut the cost of goods and remove severe impediments for larger scales. [2] The penultimate common intermediate, homobenzyl alcohol 3, features a hetero cyclic structure with three stereocenters and was built from fluo ronaphthyridines and a chiral tetrahydropyran. The two hetaryl carbaldehydes 4 and 5, both manufactured in 5-7 steps, were introduced in the final step.
