A series of first generation C-linked antifreeze glycoprotein (AFGP) analogs have been successfully prepared using conventional solid phase chemistry. These glycoconjugates range in molecular weight between 1.5 to 4.1 Kda and can be prepared using traditional linear solid phase protocol. Unlike the native system, the C-linked analogs possess enhanced chemical and biological stability and consequently are well-suited for many potential medical, industrial and commercial applications. Despite dramatic structural modifications (relative to the native system), several of these first generation analogs display significant antifreeze protein-specific activity. This implies that the rational design and synthesis of low molecular weight AFGP possessing enhanced chemical and biological stability is a feasible and worthwhile goal.
