Persistent PAR2 vasodilation in hypertensive mice

Proteinase‐Activated Receptor 2 (PAR2) caused vasodilations that were preserved in genetic hypertensive mice. In the current study we examined the effects of infusion of angiotensin II (AII) on PAR2‐mediated vascular smooth muscle relaxations and PAR2 expression on high salt diet‐induced hypertension in mice. Wire myographs were used to measure PAR2 and acetylcholine‐induced relaxations of mesenteric arteries from saline‐treated and AII‐treated mice. PAR2 relaxations persisted in the AII‐treated mice despite the reduction of acetylcholine‐induced relaxation. Blood pressures measured by radiotelemetry determined the severity of hypertension in background and PAR2 knockout mice fed high salt diets over a two week period. Baseline blood pressures and high salt diet‐induced increases in blood pressure in controls were similar to PAR2 knockout mice. We conclude that PAR2 expression did not affect blood pressure response to a high salt diet and therefore, PAR2 expression did not play a role in the blood pressure‐natriuresis relationship. Further investigation is warranted to investigate the interactions of PAR2 with endothelial dysfunction‐resistant mechanisms of vascular smooth muscle relaxation in cardiovascular diseases such as hypertension.
