Abstract

Activation of caspases is a highly recognized feature of apoptosis. Previously, we had showed that exposure of bovine oocytes to heat shock (HS) and carbonyl cyanide p-(trifluoromethoxy)phenylhydrazone (FCCP) induced apoptosis and reduced subsequent embryonic development. Experiments were performed to: 1) determine whether apoptosis mediated by caspase-9 and/or caspase-3/7 are the causes for disruption of developmental competence of oocytes during maturation induced by HS and FCCP, to depolarize the mitochondria, and 2) whether inhibition of caspases in the oocyte prevents the reduced developmental potential caused by HS. Cumulus-oocyte complexes (COCs) were matured at 39°C (control) or 41°C (HS) as well as with 0.01% (v/v) DMSO (vehicle) or 10 μM FCCP at 39°C (FCCP) for 22h. All COC groups were fertilized and cultured in vitro at 39°C for 8 days. Exposure to FCCP or HS resulted in increased activity of caspase-9 and caspase-3/7 in oocytes. Nevertheless, at the blastocyst stage only caspase-3/7 activity was increased by both treatments. Further, COCs were placed in maturation medium with vehicle (0.01% [v/v] DMSO) or 200 nM of caspase inhibitors: z-LEHD-fmk, an inhibitor of caspase-9 activity; or z-DEVD-fmk, a caspase-3/7 inhibitor; or z-VAD-fmk, a broad caspase inhibitor. COCs were matured at 39°C or 41°C, fertilized and cultured at 39°C. In the presence of all inhibitors, HS oocyte development to the blastocyst stage was similar to control and showed reduced percentage of TUNEL-positive chromatin in oocytes and blastomeres. In conclusion, HS and FCCP induced oocyte mitochondrial dysfunction by activation of caspase-9 and caspase-3/7 also caspase inhibitors rescued the capacity of oocyte to support early embryonic development.
