生命現象におけるオリゴ糖鎖の多様な働きが次々と明らかにされている。オリゴ糖の持つ生理活性をより深く理解し利用していくためには、天然型および非天然型オリゴ糖を入手する必要がある。したがって、オリゴ糖合成は化学における最も活発に研究されている分野の一つとなっている。ここで紹介するグリコシンターゼは全く新しいタイプの変異酵素であり、有機化学的な方法や糖転移酵素を用いる従来法と共に、オリゴ糖合成に利用することができる。グリコシンターゼとは、糖加水分解酵素の触媒部位の求核性アミノ酸を非求核性アミノ酸で置換した変異体のことであり、加水分解に対して不活性である。しかし、糖受容体の存在下、糖-酵素中間体に類似した構造をもつ糖供与体に作用させると、高収率で糖転移反応を進行させることができる。本総説は、筆者の研究室で開発された一連のグリコシンターゼを用いた最近の結果をまとめたものである (JACS, 1998, 120, 5583-5584)。いくつかの糖供与体をさまざまな糖受容体に転移させることを目的として、数種類の加水分解酵素に対して変異導入を行った。また、あるグリコシンターゼに関しては、糖転移反応速度の向上が図られた。このことによって、グリコシンターゼを利用する広範なオリゴ糖合成が可能となった。
