into the patients T cells.The second format is based on antibody fragments fuse with a cytotoxic T cell signaling molecule.Our laboratory isolated antibodies that recognize MHC-peptide complexes.Such TCR-like antibodies exhibit both binding properties and kinetics of antibodies (e.g.high affinity), and mimic the specificity of TCRs.We isolated a TCR-like antibody, termed F2, which recognizes an HLA-A2/wt1-derived peptide complexes expressed on the surface of leukemia cells.By Using this F2 Fab, which exhibits specificity of TCR, we were able to properly compare the biological functions of TCR-based vs. TCR-like-antibody-based engineered T cells.This comparison reveals which chimeric receptor format is superior in mediating the cytotoxic function of T cells, and may also lead to a better understanding of the mechanisms of T-cell recognition elements.
