Abstract
The first restriction enzyme, an endonuclease capable of cutting double-stranded DNA in a site-specific fashion, was isolated from Escherichia coli in 1968. Four decades later, researchers identified another system for cleaving foreign DNA—one that acts as an adaptive immune system. Conferred by clustered, regularly interspaced, short palindromic repeats (CRISPR) and a set of CRISPR-associated genes (cas), CRISPR-Cas systems differ from restriction-modification systems primarily in that they can readily acquire new specificity.
