Abstract
Glycosylation conventionally involves the reaction of a glycosyl donor with a protected acceptor having a single free hydroxy (OH) group. This chapter provides methods for selective formation of glycosidic linkages from acceptors having two or more free hydroxy (OH) groups. Intramolecular hydrogen bonding and steric effects have been invoked to explain these protective group effects on glycosyl acceptor reactivity and regioselectivity. The chapter discusses examples of the effects of donor structure and reactivity on glycosylation regiocontrol. The relative reactivity of glycosyl acceptor OH groups may vary depending on the glycosylation mechanism, which in turn depends on structural features of the glycosyl donor such as the nature of the leaving group and the protective group pattern. The ability to fine-tune or even alter the relative reactivity of OH groups by variation of protective groups, either at nearby positions on the glycosyl acceptor or on the glycosyl donor.
