058 Bradykinin B1 Receptor Antagonism Prevents Angiotensin II-Induced Erectile Dysfunction

Abstract
Chronic angiotensin II (AngII) treatment in rats induces erectile dysfunction (ED). In the penis, AngII and bradykinin have been shown to have opposing actions with AngII stimulating penile detumescence, and bradykinin promoting relaxation via the B2 receptor to facilitate erection. Bradykinin B1 receptor (B1R) stimulation offsets AngII-induced remodelling in the heart; however, the role of this B1R in erections and penile structure is unknown. The present study investigated the impact of B1R antagonism on AngII-induced ED. Sprague-Dawley rats were treated with vehicle or AngII (200ng/kg/min) in the presence or absence of the B1R antagonist R-954 (400 μg/kg/day). After 4 weeks, mean arterial pressure (MAP) was measured via carotid artery cannulation and erectile function was assessed via intracavernosal pressure following electrostimulation of the cavernous nerve. Cross-sectional area (CSA) of the coronary artery (CA), aorta, and the penile dorsal arteries were evaluated. Additionally, the left ventricle (LV) and the corpora cavernosa were analysed for collagen content via hydroxyproline assay, and protein expression of endothelial NOS, SMAD2/3, Akt, or p38.
