Abstract
Introduction and Aims: We investigated the impact of overexpression of heterogeneous nuclear ribonucleoprotein F (hnRNP F, a transcription factor) on renal angiotensinogen and transforming growth factor-beta 1 (TGF-β1) expression, tubular apoptosis and tubulointerstitial fibrosis in type 2 diabetic db/db mice. Methods: We developed and studied db/db (BKS strain) transgenic (Tg) mice specifically overexpressing hnRNP F in their renal proximal tubular cells (RPTCs). Plasma glucose, systolic blood pressure (SBP) and albuminuria were monitored weekly in adult male db/db and db/db hnRNP F-Tg mice and lean db/m littermates from 10 to 20 weeks of age. Kidneys were processed for histology and apoptosis studies. Gene and protein expression in renal proximal tubules (RPT) were evaluated by real time-qPCR and Western blotting, respectively. Urinary angiotensin 1-7 (Ang 1-7) and angiotensin II (Ang II) levels were quantified by ELISA. Rat immortalized RPTCs stably transfected with hnRNP F were also studied. Human non-diabetic and type 2 diabetic kidneys were also studied by immunohistochemistry.
