Age Dependent Variation in the Testicular Levels of Glucose Transporters (GLUTs) 4 and 8 and Their Correlation with Steroidogenesis in Mice.

Abstract
Male mammals experience decreasing reproductive function, particularly decline in steroidogenesis. Till date there is lack of information regarding relationship between metabolic changes in the testis and variation ints reproductive status. Therefore, the aim of present study was to determine the changes in glucose transport in relation to changes in testicular activity of mice from birth to senescence. The result of this study showed that there is significant variation in testicular GLUT 4 and 8 expressions during ageing. A significant correlation was observed between changes in testicular glucose level and GLUTs 4 and 8 protein levels with the circulating testosterone level and testicular StAR expression during ageing. Immunohistochemical analysis of GLUT 4 and 8 in the testis during ageing showed intense immunostaining of GLUT 4 and 8 isoforms mainly in the Leydig cells of the adult testis suggesting their relevance in steroidogenesis. The in vitro study showed a significant positive correlation between LH-induced changes in GLUT 4 (r=0.87; p<0.05) and 8 (r=0.82; p<0.05) protein levels with changes in testosterone synthesis by the testes of adult mice. Further, intra testicular 2-deoxy glucose treatment to adult mice, caused a significant decrease in testicular 3 beta HSD enzyme activity. These findings thus suggest that one of the causes of variation in reproductive status during ageing is due to variation in adequate GLUT dependent glucose transport to the testis and a sharp decline in testicular glucose level along with GLUT 4 and 8 expressions at old age might be a cause of testicular senescence.
