Organic Selenium and Vitamin B6 Synergize to Up-Regulate Glutathione Peroxidases (GPx) Expression in Dietary Supplemented Gilts.

Oxidative stress conditions occurring during peri-estrus period have been associated with reduced fertility in many species. Selenium (Se) is a mineral that participates in the synthesis of selenoproteins with strong antioxidant properties such as glutathione peroxidases (GPx). Most of the reactions involved in the fate of organic selenium for the control of the GPx system are pyridoxine (vitamin B6)-dependent. Thus, it was hypothesized here that dietary supplementation of gilts with organic Se (Se-cysteine) and vitamin B6 should enhance GPx-related antioxidant defence system in response to peri-estrus oxidative pressure. Forty-five gilts received one of the 5 dietary treatments (n = 9/group): 1) basal diet (Se = 0.2 mg/kg and B6 = 2.5 mg/kg) (C); 2) # 1 + 0.3 mg/kg inorganic Na-Se (MSe0B6); 3) # 2 + 10 mg/kg B6 (MSe10B6); 4) # 1 + 0.3 mg/kg organic Se-yeast (OSe0B6) and 5) # 4 + 10 mg/kg B6(OSe10B6). Treatments started at first pubertal estrus and lasted up to 3 d after fourth estrus. At slaughter, endometrium, liver and kidneys were collected and Se concentrations were determined. There was a clear effect (P < 0.01) of the type of dietary Se on Se content in all organs without interaction or effect of vitamin B6.; higher Se levels were observed in tissue samples from organic Se-yeast treated animals (groups 4 and 5) when compared with gilts supplemented with inorganic Na-Se (groups 2 and 3). Despite these significant variations in Se tissue content, no clear treatment effect has been observed on GPx activity determined with a standard protocol. However, quantitative RT-PCR analyses targeting different member of the GPx family (GPx1, GPx3 and GPx4) have revealed that expression of those enzymes could be increased (P < 0.01) by Se supplementation in both liver and kidneys. Strikingly, the up-regulation of mRNA expression induced by organic Se-yeast is further enhanced (P < 0.03) by the presence of vitamin B6 for all detected GPx enzymes in liver as well as for GPx1in kidneys. The extracellular GPx3 is also up-regulated by both organic Se-yeast and vitamin B6 in kidneys. Such synergistic action is not observed in inorganic Na-Se treated animals (groups 2 and 3). Additionally, while no Se treatment effect was observed in endometrium, we reported for the first time that this tissue expresses GPx2, an enzyme known to be principally located in the gastrointestinal tract. Taken together, these results highlight the regulatory role of vitamin B6 for the flow of organic Se towards the GPx antioxidant system. This unique interaction may be used to specifically increase the expression of many GPx enzymes and potentially reinforce the antioxidant defense potential in various tissues experiencing oxidative stress conditions. Such effect is highly suitable during peri-estrous period to ensure optimal ovulation and enhance the reproductive potential of the gilts.
