Identification of a Key Role for Runt-related Gene 2 in Human Cytotrophoblast Invasion.

The invasion of extravillous cytotrophoblasts (EVTs) into the underlying maternal tissues and vasculature is a key step in human placentation. The molecular mechanisms underlying the development of an invasive phenotype in EVTs include many of those first identified as having a role in cancer cell metastasis. In view of these observations, we have examined the expression, regulation and function of runt-related gene 2 (Runx2) in human trophoblastic cells in vitro, a transcription factor recently assigned an integral role in the onset and progression of cancer in a wide variety of tissues. Our initial studies using a semi-quantitative RT-PCR approach revealed that Runx2 mRNA transcripts were readily detectable in first trimester human placenta and in primary cultures of highly invasive EVTs propagated from these tissues but not in cultures of BeWo cells, a poorly invasive choriocarcinoma cell line. Similarly, interleukin-1beta and transforming growth factor-beta1, two cytokines which promote and restrain human trophoblastic cell invasion respectively, were found to differentially regulate RUNX2 expression in our primary cultures of EVTs in both a concentration- and time-dependent manner. Finally, loss-of-function studies using a siRNA strategy demonstrated that a reduction in RUNX2 expression in EVTs resulted in a concomitant decrease in the ability of these cells to invade into Matrigel Transwell inserts. Collectively, our findings demonstrate that the regulated expression of RUNX2 in EVTs confers an invasive capacity of these cells in vitro.
