Comme Tpit et NeuroDi ont un rôle essentiel à jouer dans la différenciation des cellules corticotropes, j'ai entrepris l'identification des facteurs responsables de leur activation.Dans les événements de différenciation qui impliquent NeuroDi, son expression dépend de celle d'autres facteurs 5HLH, les neurogénines (ngn).Ngn2 est exprimé dans l'hypophyse à e12,5 mais il n'est toutefois pas nécessaire à l'expression de NeuroDi.En ce qui concerne Tpit, j'ai déterminé la structure de son ADNc ainsi que le site d'initiation de la transcription, ce qui est la première étape visant à caractériser les éléments régulateurs de son promoteur.À l'échelle moléculaire, Tpit lie le promoteur de la POMC sur un site adjacent à celui de Pitxl et ces deux facteurs dépendent l'un de l'autre pour activer la transcription du gène de la POMC.En outre, Tpit et NeuroDl/Panl interagissent eux aussi de façon synergique pour activer le gène de la POMC.Puisque cette synergie dépend de Pitxl et que NeuroDl/Panl interagit avec Pitxl, ceci suggère que Tpit, Pitxl et NeuroDl/Panl forment un complexe transcriptionnel histo spécifique requis pour l'expression de la POMC.Ce complexe pourrait être le point de convergence de signaux et/ou de voies de détermination qu'il reste à identifier pour la différenciation des corticotropes à e12,5.
