Abstract
失眠症在当今社会发生普遍,现代分子学已发现失眠症可能与γ-氨基丁酸A（GABAA）受体机制、细胞因子调节机制、兴奋性氨基酸机制以及-羟色胺（5-HT）受体机制调节有关,但这些独立的机制与病理表征之间的联系仍不明确。而半夏厚朴汤作为治疗失眠抑郁的名方,其治疗机制尚不明确。为进一步探索半夏厚朴汤在治疗失眠过程中对已知或未知生物通路的作用,该研究采用基于^1H-NMR技术的代谢组学研究方法,观察给予半夏厚朴汤提取物后对戊巴比妥钠诱导大鼠睡眠实验中,大鼠体内小分子代谢物的变化并对其机制进行探讨。通过采集大鼠尿液和血清进行核磁数据检测,利用主成分分析法（principal component analysis,PCA）对大鼠尿液和血清内源性小分子代谢产物进行分析处理。并运用Simca-p 17.0（Umetrics,Umea,Sweden）和Chenomx NMR Suite 7.1（Chenomx,Inc.,Edmonton,Alberta,Canada）软件对核磁共振氢谱和相关代谢产物进行鉴定和分类。结果表明,半夏厚朴汤组和茚地普隆组与空白组区别明显,载荷图中显示最大变异代谢物,发现10个代谢物质在组间存在显著性差异。实验证明,半夏厚朴汤组和茚地普隆组均可延长戊巴比妥钠诱导大鼠睡眠时间,且半夏厚朴汤与戊巴比妥钠有协同作用。经过代谢物差异性对比,推测半夏厚朴汤对戊巴比妥钠诱导大鼠睡眠实验机制可能是通过调节谷氨酰胺、磷酸肌酸、2-酮戊二酸的含量,减轻脑内神经兴奋性从而起到调节睡眠的作用。
