Abstract
Protcin and messengeT RNA (mRNA) copy numbers vary from cell to ce]] in isogenic bacterial popu]ations.However, these mo]ecutes often exist in low copy numbers and are difficult to detect in sing:e cells.We carricd out quantitative system-wide analyses of protein and mRNA expression in indivjdual ee]ls with single-molecule sensitivity using a newty constructed yeS]ew fluorescent protein fusion library for Escherichia coli.We found that almost all protein number distTibutions can be dcscribed by the gamma distribution with two fitting paramcters which, at low expression levels, have clear physjcaj interpretations as the transcription rate and protein burst size.At high expression levels, the distributions are dominated by extrinsic noise.We found that a sing]e cel['s protein and mRNA copy numbersfoT any given gene are uncorrelated, In the symposium, we wM present our recent challenges on mRNA dynamics, in addition to the werk on single-ce]1 proteome and transcriptomeanatyses.
