Die Pathogenese fibrosierender Lungenerkrankungen ist ein komplexer Prozess, bei dem verschiedene Mechanismen zum Tragen kommen. Einer der Mechanismen, die zur progredienten Fibrosierung führen, ist die gestörte Balance zwischen Auf- und Abbau von extrazellulärer Matrix. Wir verwenden adenoviralen Gentransfer, um den Effekt verschiedener Zytokine und Wachstumsfaktoren in der Lunge zu untersuchen. Wir analysierten die Expression of Protease-Inhibitoren (PAI-1, TIMP-1) im Lungengewebe und korrelierten dies zu der Entwicklung progressiver Fibrose. TGFbeta1 resultiert in einer massiven und progressiven Lungenfibrose, die Expression von TIMP-1 ist stark erhöht und persistiert über einen Zeitraum von mehr als 3 Wochen. TGFbeta3 und CTGF hingegen verursachen eine moderate und reversible Fibrosierung, und sowohl TIMP-1 als auch PAI-1 sind nur vorübergehend erhöht. Dasselbe Phänomen ist zu beobachten in unterschiedlichen Maus-Stämmen, wo fibrose-resistente Tiere eine niedrige, fibrose-sensible Mäuse aber eine hohe und persistierende Expression von TIMP-1 haben. Die Expression von TIMP-1 scheint mit der Entwicklung progredienter Lungenfibrose eng verbunden zu sein. Wir untersuchen derzeit in komplexeren Modellen, ob und wieweit TIMP-1 eine zentrale Rolle in dem Prozess spielt und möglicherweise ein neues Therapieziel sein könnte.
