Wide-field second harmonic generation (SHG) microscopy was implemented using a high power Yb laser for applications in fast imaging of live samples and for histology slide scans in digital pathology. Live imaging of drosophila larvae muscle was demonstrated for more than 60 s continuous illumination using SHG contrast at a frame rate of 10 Hz, for applications in basic muscle research. Pathological samples of human thyroid tissue could be imaged without photodamage yielding SHG contrast in the entire slide via tiling. Wide-field SHG has the potential to be used as a high throughput histopathology sample pre-diagnosis system.
