1050: The Functional and Structural Consequences of Cavernous Nerve Injury in the Rat Model are Ameliorated by Sildenafil Citrate

whether they recieved sildenafil treatment or not.The rats were sacrificed on postoperative day 14 and their penises were harvested.The responses of the cavernous smooth muscle cell strips mounted in organ chamber were tested against acetylcholine, phenylephrine, Electrical Field Stimulation(EFS) and sodium nitroprusside (SNP).The effect of sildenafil citrate treatment on these responses were also investigated.RESULTS: The penile weight of the bilaterally denervated group was significantly lower than sham group.Sildenafil treatment had positive effects on penile weight of both bilaterally (p= 0,003) and unilaterally (p=0,004) denervated groups.In bilaterally denervated group the responses to acetylcholine, phenylephrine, EFS and SNP decreased significantly compared to sham group.In bilaterally denervated group,sildenafil treatment corrected the responses to acetylcholine, phenylephrine and SNP but there was no change in response to EFS compared to sham group.In unilaterally denervated group only the phenylephrine responses were higher and sildenafil treatment normalised these responses.CONCLUSIONS: Bilateral denervation in rats caused a denervation supersensitivity in cholinergic and adrenergic systems.Bilateral denervation may cause smooth muscle cell damage.Unilateral denervation did not seem to cause any change in cellnlar systems but it caused increased contractile responses and sildenafil treatment corrected these responses.In conclusion,sildenafil citrate treatment was shown to protect cavernous tissue against these changes in both bilaterally and unilaterally denervated rats Source of Funding: None 1050
