Abstract Wirkstoffe, die ihre Wirkung über die Ausbildung einer kovalenten Bindung entfalten, repräsentieren einen beachtlichen Anteil unseres Arsenals an effektiven Arzneistoffen. Allerdings wurde die aufwendige Entwicklung kovalenter Inhibitoren wegen Sicherheitsbedenken bis vor kurzem nur als eine wenig attraktive Strategie im rationalen Wirkstoffdesign angesehen. Die kürzliche Zulassung von vier kovalenten Kinaseinhibitoren und die Entwicklung hochpotenter kovalenter Kinasehemmstoffe mit bemerkenswerter Selektivität erhöhte das Interesse in der industriellen und akademischen Forschung maßgeblich und bestätigte das Konzept der kovalenten Kinasehemmung für klinische Anwendungen. Die Häufigkeit von Cysteinen an verschiedenen Positionen innerhalb und in der Nähe des aktiven Zentrums legt nahe, dass ein wesentlicher Teil der Kinasen durch kovalente Inhibitoren adressiert werden kann. In diesem Aufsatz geben wir eine Übersicht über die neuesten Entwicklungen in diesem schnell wachsenden Gebiet und heben die einzigartigen Möglichkeiten und Herausforderungen dieser Strategie hervor.
