Abstract
Introduction: Increasing the production of nitric oxide (NO) and subsequent toxic peroxynitrite (ONOO-), as well as increasing activity of matrix metallinoproteinase-2 (MMP-2) have deleterious effects on the myocardium. Similarly, increased phosphorylation of contractile proteins is reported in hearts undergoing ischemia/reperfusion (I/R) injury. While this phosphorylation is an important part of the normal regulatory mechanism of heart contractility, in the setting of I/R injury, the increased phosphorylation and nitration/nitrosylation of myosin light chain (MLC), increases MLC affinity to MMP-2, resulting in MLC degradation. Purpose: The purpose of this study was to protect rat hearts from injury during I/R by co-administration of subthreshold or lower concentrations of doxycycline (Doxy - MMP inhibitor), ML-7 (myosin light chain kinase (MLCK) inhibitor) and L-NAME (inhibitor of nitric oxide (NO) synthase. Methods: Hearts extracted from anesthetized Wistar rats were perfused in using the Langendorff model. The hearts were perfused for 25 min in aerobic conditions and subjected to 20 min of global no flow ischemia followed by 30 min of reperfusion. Drugs with inhibitory properties were infused 10 min before onset of ischemia and infusion of drug was stopped after 10 min of reperfusion. In addition to hemodynamic parameters, MMP-2 activity and levels of LDH and MLC1 were measured.
