Radioresistencia en glioblastoma: papel de la hipoxia en la genotoxicidad inducida por radiaciones ionizantes

Abstract

Objetivo: El glioblastoma multiforme es el astrocitoma grado IV más frecuente en la edad adulta, caracterizado una alta radioresistencia, que se asocia a la presencia de regiones hipóxicas en el tumor. Dada la importancia de la hipoxia sobre la respuesta celular a las radiaciones ionizantes, en este trabajo se estudió la relación entre la hipoxia, el daño en el ADN y la supervivencia celular. Metodología: Se utilizaron las líneas celulares de glioblastoma (U87) y cáncer de mama (MCF-7). Se evaluó el efecto de la radiación ionizante sobre la supervivencia de ambas líneas celulares en condiciones de hipoxia o normoxia, empleando el ensayo de formación de colonias. El daño del ADN se evaluó mediante la técnica de electroforesis en gel de célula única o ensayo de cometa. Resultados: La línea U87 presentó mayor radioresistencia en condiciones de hipoxia a 8 Gy, pero menor radioresistencia a dosis intermedias (6 Gy). MCF-7 presenta mayor radioresistencia en normoxia, aunque no estadísticamente significativa. En el ensayo del cometa, MCF-7 presenta mayor daño del ADN al aumentar la dosis de irradiación en hipoxia, y U87 muestra un aumento en el daño en hipoxia, que no es dosis dependiente. Conclusión: Se evidenció que en la línea U87, no hay correlación entre el aumento en el daño enel ADN en hipoxia y la supervivencia celular, sugiriendo una alta capacidad de reparación del ADN para esta línea. Contrariamente, en la línea celular MCF-7, se evidencia que sus mecanismos de reparación son más eficientes en condiciones de normoxia.
